چکیده

This article provides detailed information on manually tracked cap mesenchyme cells from timelapse imaging of multiple ex vivo embryonic mouse kidneys. Cells were imaged for up to 18 h at 15 or 20 min intervals, and multiple cell divisions were tracked. Positional data is supplemented with a range of information including the relative location of the closest ureteric tip and a correction for drift due to bulk movement and tip growth. A subset of tracks were annotated to indicate the presence of processes attached to the ureteric epithelium. The calculations used for drift correction are described, as are the main methods used in the analysis of this data for the purpose of describing cap cell motility. The outcomes of this analysis are discussed in "Cap mesenchyme cell swarming during kidney development is influenced by attraction, repulsion, and adhesion to the ureteric tip" (A.N. Combes, J.G. Lefevre, S. Wilson, N.A. Hamilton, M.H. Little, 2016) [1].

Fat4/Dchs1 signaling between stromal and cap mesenchyme cells influences nephrogenesis and ureteric bud branching.

Formation of the kidney requires reciprocal signaling among the ureteric tubules, cap mesenchyme and surrounding stromal mesenchyme to orchestrate complex morphogenetic events. The protocadherin Fat4 influences signaling from stromal to cap mesenchyme cells to regulate their differentiation into nephrons. Here, we characterize the role of a putative binding partner of Fat4, the protocadherin Dc...

CIGARETTE SMOKE/ETHANOL-INDUCED LIMB DEFECTS IN MOUSE EMBRYOS

We have produced a spectrum of limb defects in developing mouse embryos by exposing the mother to smoke from cigarettes with different nicotine concentrations, ethanol, and a combination of ethanol and cigarette smoke. The critical time of exposure was determined to be during day 10 of gestation (vaginal plug=day 0). This time is prior to the critical events which occur between the apical e...

Osr1 acts downstream of and interacts synergistically with Six2 to maintain nephron progenitor cells during kidney organogenesis.

Mammalian kidney organogenesis involves reciprocal epithelial-mesenchymal interactions that drive iterative cycles of nephron formation. Recent studies have demonstrated that the Six2 transcription factor acts cell autonomously to maintain nephron progenitor cells, whereas canonical Wnt signaling induces nephron differentiation. How Six2 maintains the nephron progenitor cells against Wnt-direct...

Distribution of Specific Glycoconjugates in Early Mouse Embryonic Notochord and Paraxial Mesenchyme

It is well known that glycoconjugate components of the cell surface and extracellular matrix, play an essential role(s) in many developmental phenomena such as cell differentiation, migration, and cellular interactions. The purpose of this study was to investigate distribution of this macromolecules during differentiation of the notochord and paraxial mesoderm. Formalin fixed paraffin sections ...

BMP4 rescues a non-cell-autonomous function of Msx1 in tooth development.

The development of many organs depends on sequential epithelial-mesenchymal interactions, and the developing tooth germ provides a powerful model for elucidating the nature of these inductive tissue interactions. In Msx1-deficient mice, tooth development arrests at the bud stage when Msx1 is required for the expression of Bmp4 and Fgf3 in the dental mesenchyme (Bei, M. and Maas, R. (1998) Devel...
